The VD16 records all the hitherto known printed works of the 16th century (1501-1600) published in the German speaking area. Broadsides, maps, and musica practica, however, have not been included.

The bibliographic cataloguing of these works began in 1969 when the Bayerische Staatsbibliothek co-operated with the Herzog August Bibliothek Wolfenbuettel; since 1990 the Landes- und Forschungsbibliothek Gotha (today: Universitäts- und Forschungsbibliothek Erfurt/Gotha) has also been a partner. It was financially supported by the German Research Foundation from 1969 to 1999. Amounting to 22 volumes and two registers, the publication of the printed bibliography came to an end in 2000.

The VD 16 is also available online in the form of a database by means of which the entire VD16 data (bibliographic records listed both in the basic work and the unpublished supplement) can be researched. The VD 16 database is continuously updated with holdings records of libraries in Germany and abroad. If necessary, existing descriptive catalogue entries (included in the printed version of the VD16) are corrected; the users are informed about these changes by the remark "Berichtigte Aufnahmen". In addition, new title records are submitted by German and foreign libraries on a regular basis. The database currently consists of some 110.000 catalogue entries, of which there are some 27.000 new titles from the supplement (with a "ZV" number).

The holdings of the Bayerische Staatsbibliothek pertaining to the VD16 have been digitised, with the financial support of the German Research Foundation, since 2006. According to their funding guideline "Digitalisierung der in nationalen Verzeichnissen nachgewiesenen Drucke - VD16/VD17" the VD16 is considered as the basis for other coordinated digitisation projects.
As yet (April 2012), some 30 per cent of the sum total of VD16 catalogue entries have been enriched by links to the respective comprehensive digital copies.
